    Abstract:
    Advantage spoilage bacteria were identified in chilled duck by traditional bacteria isolation method and 16S rDNA flora analysis. 17 advantage spoilage bacteria had been detected in chilled duck by traditional bacteria solation method. 16S rDNA flora analysis showed that dominant spoilage bacteria in chilled ducks was of Pseudomonas, being of 54%. Strains of Aeromonas and Enterobacter were the second groups accounted for 15%. The content of a strans of Lactobacillus, Arthrobacter or Bacillus, as well as an unidentified strains,was accounted for 4%.

    Abstract:
    This study aimed to investigate the effect of Yunnan wild elsholtzia oil on learning and memory function of mouse offspring . From the first day of pregnancy, Kunming mice were divided into five groups: blank control group, positive control group, low-dose, medium-dose and high-dose groups and were intragastrically administrated with the elsholtzia oil gavage till being weaned. According to intragastic administration dose for the pregnant rats, the weanedmale mouse was intragastically administrated till 49 days after birth and then tested for the Morris water maze test. The fatty acid contents of the liver and brain organization were detected. Compared to the blank control group, navigation test showed that each group were statistically significant (p<0.01) in the 5 th day. Space exploration experiment showed that only the positive control group was statistically significant (p<0.05). By analysis of TPro, NO, AchE , it was found that he positive control group, medium-dose and high-dose groups were statistically significant (P<0.05). The contents of the fatty acids in liver of the positive control group and the high-dose group were statistically significant (p<0.05). Contents of oleic acid and ALA in high-dose group were statistically significant (p<0.05) in brain tissue. ALA-riched eElsholtzia oil can partly enhance the learning and memory in mouse offspring mouse., due to the hihger TPro and ALA contents but lower NO and AchE contents by feeding ALA-riched elsholtzia oil.

    Abstract:
    In order to probe an effective way of cloning the endogenous inductive promoter and isolating inductive differentially-expressed gene fragments from Yeast-like conidia of Tremella fuciformis, conidia was cultured by two carbon source inducers(ethanol and cellobiose). The inductive differentially-expressed gene fragments were isolated by cDNA-AFLP from the two kinds of differential cells after the growth status analysis. Result showed that, the dehydrogenase activity of the cells cultured with ethanol was 168% higher than that with glucose. The maximum growth rate of the cells cultured with cellobiose reduced by 32.2%. 14 differentially-expressed gene fragments were isolated from the tow differential cells 5 of these gene fragments expressed separately in the cells cultivated with ethanol and cellobiose, which were homologous with the sequence Codes of cytochrome C oxidase and cytochrome b. The gene expression was related to ethanol metabolic and stress induction. This research mayb be of benefit to construction of edible fungus inducible expression system and highly effective bioreactor of Tremella fuciformis.

    Abstract:
    A Blumlein type pulse forming network system was developed for treating Vibrio parahaemolyticus suspension solution. The results showed a high disinfection ability of this high voltage pulse electric fields on Vibrio parahaemolyticus. Meanwhile, the cytotoxicity of the bacteria was reduced after the bacteria were treated by this system. The expression level of three type secretion system -related genes increased with the increase of voltage, indicating that high-voltage pulse electric fields affected the pathogenic of bacteria. This research could be a theoretical base for liquid food disinfection, bioproducts and vaccine inactivation.

    Abstract:
    The migration of formaldehyde in melamineware were analyzed by investigating the influence of different marinating solutions, temperature, time, cutting size, microware treatment on the migration quality of formaldehyde from melamineware. Higher quality of migrated formaldehyde was found in the acid solution than in other solutions. Migration quality of formaldehyde rose abruptly above 80 ℃. A linearity relationship between soaking-treatment time and migration quality of formaldehyde was found. In addition, decreasing the granularity of melanmineware or increasing the microwave-treatment time (above 15 min) led to higher content of the migrated formaldehyde.

    Abstract:
    Resveratrol is a grape polyphenol with a variety of bioactivities.The precursor substances of resveratrol, polydatin, have high content in plants, such as Polygonum cuspidatum. In this paper, screening medium with polydatin was used to screen a strain that can convert polydatin into resveratrol from Polygonum cuspidatum. Then the screened strain was identified by morphology and 18S rDNA sequence analysis methods. The identified results showed that the strain was Penicillium camembert.

    Abstract:
    The properties of corn starch-based edible films formed from cross-linked esterified, esterified cross-linked modified starch were concerned in this paper. The crystallization degree and microstructure of the starch and films were studied by Fourier transform infrared spectrometer (FT-IR), X-ray diffraction and scanning electron microscopy respectively. Applications in preservation of litchi and clausena lansium were tested. The results showed that tensile strength of cross-linked esterified films were increased apparently while the water vapor permeability of esterified cross-linked films were decreased. Light transmittance of the two kinds of film was improved, but the preservation effects on litchi and clausena lansium were not significant.

    Abstract:
    Six Gram-positive bacteria with strong acidogenicity were separated and purified from fresh feces of healthy babies, and their bacteriostasis in vitro were tested. Three strains with high inhibition effect on diarrhoeic Escherichia coli (O78, O88 and O149) were screened. The results of biochemical identification and 16S rRNA comparison analysis showed that all the three screened strains were Lactobacillus casei. The challenged test in mice and chickens by one strain with strongest inhibition on pathogenic E. coli showed that the strain was safe, nontoxic and appropriately improved the animal feed conversion rate, which can used for prevention and treatment of diarrhea effect.

    Abstract:
    ζ-Carotene desaturase (ZDS) is one of the key enzymes in the carotenoid biosynthetic pathway. According to the full-length sequence of the ZDS cDNA in D. bardawil that our laboratory has obtained before, the coding region sequence of ZDS were isolated by the approach of PCR and then flanking sequence of the coding region were amplified by the method of genome walking, including promoter and terminator of ZDS, followed by analysis of these sequences by bioinformatics tools. The whole sequence of ZDS was11896 bp. It contained 6435 bp coding region (from ATG to TAA), 4091 bp upstream of the coding region, and 1370 downstream of the coding region. Blastn the coding region and ORF of ZDS showed that there were 12 exons and 11 introns in coding region. The total length of the introns was 4086 bp, about 2.68 fold of the total length of exons (1749bp). Each intron started with GT and end with AG, belonging to the most common style of exon. By analyzing the promoter of ZDS gene, we found that there are many transcription factor binding sites, including “GAGA box”, ASF1, related with plant light regulation; “ACGT box”, required for etiolation-induced expression of early responsive to dehydration.

    Abstract:
    To explore the application of gramma irradiation and additives in Fresh-keeping of the light salted Pseudosciaena crocea, the influence of low-dose gamma irradiation(1 kGy) and additives (0.3% sodiumlactate +0.5% citric acid )on quality and shelf life of mildly salted Pseudosciaena crocea stored at 25℃was discussed by analyzing the changes in sensory quality, microbiological and chemical properties. The results showed that both of the two processing methods can remarkably prolonged the shelf life. Compared to the shelf life of control group(9 days), the shelf lifes of experimental groups can be extended to 16 days and 13days, respectively. 1 kGy gamma rays had better preservation effect than additives (0.3% sodiumlactate +0.5% citric acid). The work can provide technical references for industrial application.

    Abstract:
    The rice flour was modified by acid and heat-moisture treatment in the experimental condition in this paper. Polarization microscope, scanning electron micrograph, infrared spectrometer, X-ray diffraction (XRD), differential scanning calorimetry (DSC) instrument, Brabender viscosity instrument were used to study the properties of rice flour before and after the treatment. The results indicated that its protein content was reduced, while other components kept constant. The processed rice flour granules were damaged lightly by acid and heat-moisture. X-ray diffraction analysis showed that the acid and heat-moisture treatment made crystalline structure of starch reduced. No clear change was observed in infrared spectrometer. DSC showed that the endothermic peak of rice starch moved backward, and the absorption enthalpy (△H) decreased. Brabender paste viscosity curve showed that the samples had low viscosity. The in-vitro classification of nutritional segments of modified rice flour showed that its slowly digestible starch (SDS) content increased to some extent.

    Abstract:
    The main taste-active components in the microwave-treated/boiled Litopenaeus vannamei, including the free amino acid,nucleotide, organic acid, betaine and inorganic ions, were quantified with instrumental analysis methods. The free amino acids contents in the microwave-cooked shrimp and boiled shrimp were decreased by 15.81% and 20.19%, respectively. After microwave and water-boiled treatments, the contents of nucleotides were decreased by 25.07% and 25.89%, respectively, while adenosine monophosphate (AMP) were increased by 4.01% and 5.95%. The contents of lactic acid and betaine were significant difference, lactic acid were decreased by 53.64% and 77.24%, respectively, while betaine were 7.26% and 8.83%, respectively. Moreover, the contents of inorganic ions in the microwave-cooked shrimp were higher than those of the boiled one. Expect the AMP, higher free amino acid,organic acid, betaine and inorganic ions contents were found in the microwave-cooked shrimp than in the boiled shrimp,,showeing that the taste of the microwave cooking shrimp was better than the boiled shrimp.

    Abstract:
    In this experiment the free radical-scavenging activity of various components from terminalia chebula ritz was studied. The results showed that all extracts of terminalia chebula ritz stems had strong DPPH? scavenging effects. The activity-decreased sequence of the extracts by different extracting regents was: methanol>ethyl acetate>acetone>ether>petro-ether. The methanol extract (1mg/mL) had better activity than that of TBHQ. The activities of the component A was 1.6 folds higher than the original. The chromatographically purified component C showed the strongest activity among all samples. Its activity was a little stronger than that of tea polyphenol, as 1.2 folds higher than that of TBHQ. By color test, UV-Vis and H1 NMR analysis, all of the purified components were confirmed to be the derivatives of myrobalani-tannin or their hydrolyzed segments.

    Abstract:
    The change of the aerobic plat bacteria count in high salinity fish sauce during storage and influence of different salt content on its quality were studied. Results showed that the aerobic bacteria count reduced gradually during the storage time. And the same phenomenon occurred in three grades of fish sauce in one month. High salinity had a significant effect on bacteriostasis of fish sauce as the storage time extended. In relatively, the aerobic plate count of the 25% salinity were decrease by 97.91% and 99.73% respectively then of the 10% salinity when storage at 15 d and 30 d. 25% of salinity could well maintain the quality of fish sauce during storage.

    Abstract:
    The food-grade Nisin controlled expression (NICE) system of Lactococcus lactis (L.lacti) was studied emphatically as a vector of recombinant protein expression. The therapeutic proteins or protective antigen protein expressed can secrete and anchor in the cell envelope or bacterial peptidoglycans and thus the NICE system of L.lacti can present the vaccine antigen to the mucosal surface and result in a specific s-IgA response. Using recombinant L.lacti that present antigens as live mucosal vaccines provides an effective way to prevent microorganisms invading and shows a promising future. In this study, BALB/c mice were immunized with the bacterial supernatant to evaluate the mucosal immune response and Immune tolerance of recombinant L.1acti PNZ8149/NZ3900-M/PRRS strain that expresses ORF6 gene of PRRSV,. IgG in serum and s-IgA in lung lavage fluid and intestinal washes were analyzed by using a similar enzyme-linked immunosorbent assay (ELISA). For evaluating changes in cytokines IL-4 and IL-10, the spleen collected was analyzed. The results clearly showed that mice immunized with recombinant strain PNZ8149/NZ3900-M/PRRS could induce remarkable special s-IgA in lung lavage fluid, at the whole experimental session, compared with the control group I and II. The level of special s-IgA and IgG showed significantly increase(P<0.01) and IL-4 and IL-10 contents of cytokines showed little difference to those of the control groups. All the results indicated that mice immunized by the recombinant L.lactis expressing ORF6 gene of PRRSV can significantly induce mucosal immune responses and avoid producing the phenomenon of mucosal immune tolerance.The study was expected to lay a theoretical foundation on developing of the gene engineering L.1acti s mucosal vaccines.

    Abstract:
    In this paper, the effect of nutrition solution on the freshness of vegetable was researched, and the concentrations of magnesium and ferric in vegetables were determined. The results showed that the nutrition solution could evidently prolong the freshness period. By this treatment, pakchoi cabbage and cauliflower could be freshly stored for 25 days and 15 days, respectively. The concentrations of vitamin C and titratable acids of the treated vegetables were higher than those of the controls, while the nitrite concentration was lower than that of the control group. The determination of trace elements showed that the vegetable could still absorb the nutrition elements after picking.

    Abstract:
    Two Aspergillus strains (Aspergillus soji, A. orgzae) were used in this study for 6-day solid-state fermentation of rice at35°C and humidity >90%. The metal chelating, polyphenol oxidase inhibitory activity, oxygen radical absorbance capacity, and total phenolic contents of fermented rice during fermentation were analyzed. The rice fermented by A. orgzae had higher metal chelating, polyphenol oxidase (PPO) inhibitory activity and oxygen radical absorbance capacity (ORAC) than that fermented by A. soji. In terms of total phenol content, a similar fermentative effect was observed for both of the strains. In addition, these four properties of fermented rice tended to be stable at the fourth day of fermentation. Therefore, rice fermented by A. orgzae for four days was selected for the evaluation on anti-tyrosinase activity. Lineweaver-Burk Plot was adopted to investigate the inhibitory effects of bioactive compounds in fermented rice by A. orgzae on the oxidation of L-DOPA catalyzed by mushroom tyrosinase (M-PPO). The potential mechanism of the anti-tyrosinase activity was estimated based on the results of Lineweaver-Burk Plot. Moreover, the experiments on anti-melanosis of shrimp hemolymph and shrimp further supported the effective anti-tyrosinase activity of fermented rice by Aspergillus.

    The casting solution for forming Al2O3/ PVDF hybrid membrane was prepared using PVDF modified by KOH in the solution of methanol as a membrane-forming polymer, using vinyl-trimethoxy-silicane as a coupling agent and using aluminium isopropoxide (AIP) as an inorganic precursor.,Al2O3/PVDF hybrid membrane linking with chemical bond was obtained with the dry-wet inversion method. The effects of AIP on the phase behavior and gelation kinetics of the casting solution were studied so as to explore the mechanism of the influence of AIP on micro-structure of the Al2O3/PVDF hybrid membrane. Results showed that the change in the phase equilibrium of casting solution was encouraged by an increase in AIP concentration and casting solution became thermodynamic unstable, so that compatibility of the casting solution with non-solvent was decreased and the gelation rate of casting solution was accelerated. The highest gelation rate was found in the initial period of membrane formation, and then it slowed down. The gelation rates for different casting solution varied with different casting solutions. Both gelation rate and phase separation rate for the casting solutions were improved by the increases of AIP concentrations; however the gelation rate decreased when AIP concentration reached 20%.

    Abstract:
    The conditions for solid fermentation of a strain of Pecillium citrinum GX-K with high yield of nuclease P1 were studied. Wheat bran was US as the substrate of solid fermentation and the other optimum fermentation conditions were: 4% of sucrose, substrate-water ratio 1:0.9, fermentation temperature 30 oC and fermentation time 72 h. The activity of nuclease P1 under the optimum fermentation conditions reached 5783.41 U/g.

    Abstract:
    In this paper, the enzymatic phosphorylation of inosine in the 5′-position to produce inosine-5′-monophosphate (5′-IMP) with whole cells was studied, using the engineered E. coli BL21 with the expression of acid phosphotransferase (PHOC) phoc gene originated E.aerogenes IAM 1183. A maximum conversion rate of 63.36% was obtained after 6 h of enzymatic reaction at 30 oC in 100 mM inosine and 120 mM pyrophosphate (ppi) with PhOC activity of 45 U in a 150 mL reaction system. To further improve the conversion rate, the effects of three surfactants on the conversion rate were studied, and the conversion rate of inosine was increased to 74.59% at the addition of 0.3% Trion-x-100. This study provided basic data for the industrial application of the PHOC in production of 5′-IMP.

    Abstract:
    The content of proteins and the enzymatic hydrolysis effects on different kinds of trash fishes were compared. And the enzymatic hydrolysis effects of Hanergula zunasi and Clupanodon punctatus were be modified. The results showed that the protein contents of Decapterus maruadsi and Hanergula zunasi are more than 17%, and for Clupanodon punctatus, Setighnna taty,Hanergula zunasi,Mugil cephalus and Llisha elongata,the protein contents were ranged in 15-16%. The enzymatic hydrolysis effects of Decapterus maruadsi, Setighnna taty, Hanergula zunasi and Mugil cephalus were better than those of Hanergula zunasi, Clupanodon punctatus and Llisha elongate. The best enzymes for Hanergula zunasi hydrolysis are the mixture of 0.05% papain, 0.1% Flavourzyme, 0.05% Alcalase and 0.15% trypsin, with which 86.07% of protein recovery, 49.24% of degree ofhydrolysis and 11.30 ml of the production quantity were achieved. The best enzymes for Clupanodon punctatus were the mixute of 0.15% papain, 0.1% Flavourzyme, 0.05% Alcalase and 0.15% trypsin, with which 83.22% of protein recovery, 49.14% of degree ofhydrolysis and 10.91 ml of the production quantity were achieved.

    Abstract:
    Response surface methodology was applied to analyze the effect of carbonation conditions including of litchi juice syrup content, carbonation time and pressure on sensory scores of sweetness, acidity, carbonation, flavour and overall acceptability for carbonated litchi beverages. The conditions of preparation were established by means of a Box-Behnken for litchi juice syrup content, carbonation time and pressure in the range of 20%~40% (V/V), 5-15min and 0.5~0.23 MPa, respectively. The response variables were fitted to models using multiple regression analysis. The results showed that the determination coefficient R2 of models was 0.970, 0.855, 0.977, 0.949 and 0.899 for Y1 (sweetness), Y2 (acidity), Y3 (carbonation), Y4 (flavor) and Y5 (overall acceptability), respectively. By using the approach of desirability function, the optimum carbonation conditions were found to obtain desirable scores for all these sensory attributes simultaneously, which were: the litchi juice syrup content of 28%~30% (V/V), carbonation time of 12~15 min and carbonation pressure of 0.21~0.23 MPa. Under these conditions, the scores from the verification experiments were 3.1 for sweetness, 3.0 for acidity, 3.5 for carbonation, 4.0 for flavour and 4.0 for overall acceptability, which were in good agreement with the predicted values.

    Abstract:
    Cauliflower foliage was used as the raw material for extracting chlorophyll with microwave extraction. Some factors were considered for single factor experiment,such as size of cauliflower foliage powder, density of cell wall crack agent, microwave power, irradiation time, extractant concentration,solid-liquid ratio. L16(45) orthogonal experiment was adopted for getting the optimal mixed test conditions. The result showed that the extraction ratio of chlorophyll could be achieved as 6.843mg/g under the following conditions:60% of density of cell wall crack agent, 30s of irradiation time, 90% of extractant concentration and 1:20 of solid-liquid ratio. Compared with the traditional technology, microwave assisted extraction method can increase extracting rate of chlorophyll by 62.2%.

    Abstract:
    Strains of probiotic bacteria which were isolated from dairy products, health products and internationally-recognized probiotics were chose as test strains. Through the domestication, their characteristics were studied by evaluating the curd, curd time, sensory quality, viable count of probiotics, and pH value of the fermented mungbean milk to obtain several strains with high reproductively ability and fermentation activity .The result indicates that Lactobacillus casei 07-211, Lactobaccillus helveticus AS 1.1877T, 05-29 and Lactobacillus plantarum AS.1.2986, 07-191 were screened from 13 strains of lactic acid bacteria for mungbean milk fermentation. After a 6-hour fermentation of the mungbean milk fragrance with those three strains, curd could be formed with, the viable cell number reached 3.9×108, 1.6×108 and 1.4×108 cfu/ml, respectively. The pH values of the fermented soybean milk by these three strains were 4.16, 4.30 and 4.32, respectively, and the acidity values reached 83.5, 81.0 and 75.2 °T, respectively. This research can provide reference for development of functionnal probiotics yoghurt.

    Abstract:
    The effects of calcium chloride amount, curd enzyme quantity, soybean milk amount and curd temperature on a camembert cheese preparation were studied. According to Box-Behnken center combination experimental design principle, second multiple regression model were estibulshed using curd effect as response value and the effectiveness of the model was verified. The results showed that the best cheese production process conditions were as follows: 0.06% calcium chloride, 0.02% curd enzyme, curd temperature 32 ℃ and row milk time 30 min, under which the estimated value and verified yield values of curd effect were as high as 89.63 and 89.50, respectively.

    Effects of the starch phosphate monoester on the steamed dumplings wrappers were investigated by orthogonal experimental design. The results showed that the starch phosphate monoester was the most significantly influential factor fo the quality of steamed dumplings wrappers, followed by water addition and water temperature. The optimal technical parameters for preparation of steamed dumplings wrappers were phosphate monoester starch 7% (count as wheat flour), water temperature 95℃, and water addition65%(count as wheat flour).

    Kudzu vine Root dietary fiber was used to made functional biscuits. Results showed that the best recipes were: flour 100 g, Kudzu Vine Root dietary fiber 30%, oil 10%, bulking agent 1% [ratio of sodium bicarbonate and ammonium bicarbonate 1:0.5 (m/m)] and sugar 30%. The biscuits were crispy and had attractive colors and unique flavors.

    Abstract:
    A monoclonal antibody (MAb) against norfloxacin was prepared using hybridom a techniques, and indirect competitive Fluorescence immunoassays for norfloxacin (NFLX) was established successfully. The regression equation was I=31.92logC-5.0083 (r= 0.9965) with the limit of detection about 6.09 μg/L. The linear detection was ranged well from 10μg/L to 500μg/L. The cross-reaction tests indicated that most structurally related phenolic compounds did not interfere with the analysis of NFLX. The anti- NFLX antibody had excellent recognition specificity to NFLX. The result developed in this study indicates this method would practicable to detection norfloxacin residues in animal derived food.

    Samples were treated by wet microwave digestion procedure with HNO3/H2O2 and the mineral elements in bee-pollen of Brassica campestris L. were completely analyzed by inductively coupled plasma optical emission spectrometer (ICP-OES). The results showed there were 17 mineral elements in the samples. The RSD of the determined values were between 0.85% and 6.97%. And RSD of the contntes of14 elements was within 5%. Standard recovery rate of 13 elements was 93.5 %-107.3 % and 9 mineral elements was within 100 %±5 %. The mass fractions of such necessary macroelements, including S, P, K, Ca, Mg and Na, were 0.74%, 0.59%, 0.35%, 0.23%, 0.13% and 0.0064%, respectively. The contents of the microelement Fe, B, Zn, Mn, Se, Cu, Sr, Ba, Ti, Si and Al were 14.3, 6.41, 2.40, 1.64, 1.23, 0.69, 0.54, 0.44, 0.10, 50.7 and 25.8 mg/100g, respectively.

    Fifteen honey samples which found to contain no protein from 120 specimens were further investigated by element analyzer-isotope ratio mass spectrometry, liquid chromatography-isotope ratio mass spectrometry and liquid chromatography, the protein components in the honey samples were detected. It was found that adulteration existed in all the tested fifteen samples. The samples were found to be adulterated by C-4 plant hydrolyzate, C-3 plant hydrolyzate or C-3 plant high fructose corn syrup.

    A high performance liquid chromatography tandem quadrupole mass spectrum (LC-MS/MS) method was developed for confirmatively determining of benalaxyl residues in 13 foodstuffs. The benalaxyl was extracted with acetone-hexane in animal-origin foods and ethyl acetate in plant derived food. The purification was used with Carbon-NH2 solid-phase extraction (SPE) column. A mixture of acetonitrile and 0.1% formic acid was selected as the mobile phase and the separation was carried out on Shiseido MGⅡ C18 column. The compound was analyzed quantitatively by mass spectrometry under positive electrospray ionization (ESI+) multiple reaction monitoring (MRM) mode. The linear range of analyte was from 2 μg/L to 100 μg/L and the limit of quantification (LOQ S/N≥10) was 5 μg/kg. Spiked with three concentration levels in 13 matrices, the method recoveries were between 66.0% and 104.2% with relative standard deviations 4.4% to 12.3%. The validated method was accurate, rapid and suitable for qualitatively determining benalaxyl residues in many foodstuffs.

    Abstract:
    A new method for simultaneous determination of seven organic acids in soy sauce was developed with solid phase extraction – high performance liquid chromatography. The sample was purified by a Strata SAX SPE cartridge after pretreatment, then separated using a reversed-phase Agilent SB-Aq(150 mm×4.6 mm×5 μm)column. 0.01 mol/L (NH4)2HPO4 (pH 2.7) solution was used as the mobile phase at a flow rate of 0.7 ml/min. The detection was performed on an ultraviolet detector at 210 nm. The relative standard deviations were 0.13%~1.9% and the recoveries were 95.0%~105%. The correlation coefficients for the linear equations were 0.9993~0.9999. The results mentioned above indicated that the method was simple, rapid and accurate. It could be applied to the determination of organic acids in soy sauce.

    A new enzyme sensor for determination of dioleglyceride was constructed by nano-nickle oxide modified through electro- chemical oxidation sedimentary reaction on the screen-printied electrode (SPE) and lipase immobilized by chitosan. Using ferricyanide as the electron mediator, , the CV behavior of dioleglyceride on lipase-NiO-SPE were evalutaed in the solution of 1% Triton-X100 and PBS buffer (0.1 mol/L, pH 7.0). A good linear relationship was obtained between dioleglyceride ranging in 0.04~180 μmol/L with cyclic voltammetry (R =0.993) and detection limit of 0.01 μmol/L. This method was simple, rapid, accurate, and steady for the determination of dioleglyceride.

    The HPLC conditions for free amino acids detection was explored and a method for simultaneous determination of 16 kinds of free amino acids was established firstly. With this method, the free amino acids in yeast extract before and after heat treatment, HVP power and enzymatically-hydolysed pork were analyzed. It was found that yeast extract without heat treatment.had the most types and the highest contents of free amino acids. With the decreasing of reducing sugar(xylose) in heat treatment, the contents of free amino acids decreased. Yeast extract showed to be a good source of amino acids in the production of meat flavors with Maillard reaction.

    The purpose of this study was to establish an ultra-sensitive and handy headspace gas chromatographic for the determination of alcohol in rat blood by gas chromatograph. The blood samples containing alcohol and tert-butanol (internal standard, IS) were heated in a water bath. The headspace vapor was drawn into a syringe and injected into a GC port and the experimental conditions of headspace and GC were optimized by taking the peak areas as index. The results showed that the samples heated at 70 ℃for 15 min had a steady peak area and good vapor-liquid equilibrium. Under the optimized conditions, the target peak and the internal standard peak were clearly separated. In the range of 0.05~12 mg/mL, the linear relation between peak area and concentration of the alcohol was found. RSD was 1.63% by testing the standard solution for seven times. The detection limit was 0.005 mg/mL and the reclaim ratio was 93.4%~103.2% (RSD≤3.41%). In conclusion, this method was simple, flexible and effective to detect the alcohol concentration in rats’ blood and can be used widely.

    Water-soluble vitamins were determiend by Ultra-performance liquid chromatography (UPLC) equipping ultraviolet detector and fluorescence detector in series. Samples were separated by a Acquity UPLC HSS T3 (2.1×100 mm×1.8 μm) column using 0.1% trifluoroacetic acid-acetonitrile as mobile phase in gradient elution mode. Six water-soluble vitamin components in health foods were simultaneously determined, including ascorbic acid (Vc), nicotinamide (B3), thiamine (B1), pyridoxine (B6), folic acid (B11), riboflavin (B2), etc. This method was enabled to achieve wide linear range, well correlation, low detection limit and fast analysis. The results were in accordance with those wtih GB methods.
