Abstract:UV-visible spectrophotometry was used to establish the most suitable complex chromogenic system for the detection of Corn Silk Total Flavonoids (CSTF) content. In the experiment, 60% ethanol was used as solvent to extract CSTF, and CSTF was extracted by different polar solvents. Finally, n-Hexane, ethyl acetate and n-Butanol extracts were obtained. The contents of total flavonoids in ethyl acetate and n-Butanol extracts under NaNO2-Al(NO3) 3/AlCl3-NaOH and KAc-Al(NO3)3/AlCl3 complexation systems were compared with different types of flavonoids in corn silk. The content of flavonoids in ethyl acetate and n-Butanol extracts was determined by High Performance Liquid Chromatography (HPLC) to identify the most suitable reference standard and color system. The results showed that the contents of flavonoids in ethyl acetate and n-Butanol were 83.45% and 57.36%, respectively. The results showed that the KAc-Al(NO3)3 complex color system with rutin as the reference standard was the closest to the content of total flavonoids detected by HPLC. In summary, the KAc-Al(NO3)3 complex chromogenic system is the best method for detecting the total flavonoid content of corn silk and each extract. This method detects the flavonoid content of CSTF extract is 1.08%. The method is simple and accurate, which can provide a basis for the development and utilization of flavonoids in corn silk and quality control.
