    建立了一种苦荞中芦丁和槲皮素含量的测定方法。以超声法进行提取,并分别以芦丁和槲皮素为标品,采用岛津ODS柱(4.6 mm*150 mm, 5 μm),以乙腈–水(含2%的冰醋酸)为流动相,梯度洗脱,流速:1 mL/min,检测波长:360 nm。芦丁进样量在0.023~1.150 μg范围内(r=0.9996),槲皮素进样量在0.02~0.60 μg范围内(r=0.9999)线性关系良好。平均加样回收率芦丁为98.5%,槲皮素为95.8%。该方法简便、快速、准确,可测定苦荞中芦丁和槲皮素的含量。

    Abstract:

    A new method for determination of rutin and quercetin contents in buckwheat was developed in this paper. The extracts from buckwheat by Ultrasonic extraction was analyzed on a Shimadzu ODS column(4.6 mm * 150 mm, 5 μm)with a mobile phase of acetonitrile - water (containing 2% glacial acetic acid) for gradient elution at 360nm. And the flow rate was 1.0 mL/min. The linear ranges of rutin and quercetin concentrations were found within 0.023~1.150 μg (r = 0.9996) and 0.02~0.60 μg (r = 0.9999), respectively. The average recoveryies of rutin and quercetin were 98.5% and 95.8%, respectively. The method was simple, accurate and showed good reproducibility, which was suitable for the determination of rutin and quercetin contents in buckwheat.
