研究多穗柯叶醇提物(LEL)对2型糖尿病模型小鼠糖脂代谢及氧化应激反应的改善作用。将成模的2型糖尿病(T2DM)小鼠模型,分为空白对照组、模型对照组、阳性对照组及LEL高、低剂量(30、15g/kg)组,连续给药4 W。实验期间观察小鼠的毛色、精神状态并测定空腹血糖(FBG)。实验结束后,检测小鼠口服糖耐量及血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)水平,检测肝组织中肝糖原、过氧化物歧化酶( SOD)、还原型谷胱甘肽(GSH)、丙二醛( MDA)水平,并进行肝组织病理切片检查。结果显示,LEL可改善T2DM小鼠的精神状态及多尿症状。低、高剂量组较模型对照组空腹血糖分别降低了45.38%、46.25%,血清OGTT、TG、TC、LDL-C水平显著降低,HDL-C显著升高(P<0.01);肝组织中肝糖原含量及SOD、GSH活性显著升高,MDA含量显著降低(P<0.01),脂肪变性及炎性细胞浸润程度明显减轻。结果表明,LEL明显降低T2DM小鼠血糖,可能与升高肝糖原含量、改善糖质代谢与提高氧化与抗氧化水平有关。

To study the improvement effects of hypoglycemic, lipid metabolism and oxidative stress in type 2 diabetes model mice with Leaf Extracts of Lithocarpus polystachyus Rehd.(LEL). The mice were successfully induced to T2DM model,and the mice were randomly divided into following groups,normal control group,model control group,positive control group,LEL high and low-dose(30,15 g/kg)group,and mice of each groups were continuously given corresponding drugs for 4 weeks.During the experiment,the fur color,mental state,body weight,fasting blood glucose (FBG) of mice were measured.The oral glucose tolerance,liver coefficient and the serum of total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C) levels, and the liver tissue of glycogen,superoxide dismutase(SOD),glutathione(GSH), malonic dialdehyde (MDA) levels and histopathological examination of liver tissue were measured after the experiment. The results showed that LEL could improve the mental state and polyuria of T2DM mice.Compared with model control group,fasting blood glucose in low and high dose groups has decreased 45.38% and 46.25%, respectively,and the serum of OGTT, TG, TC and LDL-C levels of LEL low and high-dose groups were significantly decreased, while HDL-C was significantly increased (P<0.01).The content of liver glycogen and activities of SOD and GSH in liver tissue were significantly increased , while the content of MDA was significantly decreased (P<0.01)in the LEL low and high-dose groups.The Steatosis and inflammatory cell infiltration inT2DM were significantly reduced after giving LEL four weeks.Our results indicate that LEL can significantly reduce blood glucose in T2DM mice, which may be related to increase hepatic glycogen content, improve lipid metabolism and increase antioxidant level.
