[摘要]
本文研究黑果腺肋花楸(黑果)果实、叶子的多酚提取物对抑郁小鼠模型抑郁调节的作用机制,旨在通过饮食干预达到调节抑郁的目的。注射4 mg/mL皮质酮构建小鼠抑郁症模型,采用行为实验、16S rRNA基因测序技术等对小鼠行为、肠道菌群与脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)的变化规律等进行研究。研究表明黑果多酚提取物可以显著改善抑郁症小鼠游泳、糖水偏好等行为,其中黑果叶子提取物效果好于黑果果实提取物;肠道菌群多样性显著提高18.23%;大脑中BDNF水平可提高81.39%。黑果多酚提取物通过调控肠道菌群、大脑BDNF途径影响抑郁小鼠行为,改善抑郁状态,从而发挥抑郁调节作用,为黑果宽渠道开发提供了新思路,高值化应用提供了理论指导,以期为饮食干预调节抑郁症提供新资源,也丰富了功能性食品类型和理论。

In this paper, the anti-depression mechanism of polyphenol extracts from the fruits and leaves of Aronia melanocarpa was investigated, aiming to regulate depression through dietary intervention. The depression model of mice was established by injecting 4 mg/mL corticosterone, the changes of behaviour、intestinal flora and BDNF in mice have been studied using 16S rRNA gene sequencing technology, behavioral tests and so on. The results showed that immobility time and sucrose preference of depression mice could be improved significantly by polyphenol extract, and the effects of extract from the leaves were better than that from fruits; compared with the blank control group, the diversity of intestinal flora were significantly increased by 18.23%; after the depression mice were injected with polyphenol extract from fruits of Aronia melanocarpa, the level of BDNF could be increased by 81.39%. The polyphenol extract could improve depression by regulate intestinal flora and BDNF, thus had a protective regulation effect on the depression of mice. It provided new ideas and theoretical guidance for the high-value use of Aronia melanocarpa had been provided in this paper, so as to provided new resources for diet intervention and regulation of depression, and expanded functional food types and theories.
