针对花生致敏蛋白Ara h 2、Ara h 3及芝麻蛋白2S albumim，使用实时荧光定量PCR法（Real-time PCR）及酶联免疫吸附法（ELISA）对纯芝麻酱中上述蛋白进行检测。两种方法的特异性、重复性方法学验证显示均良好。验证后使用实时荧光定量PCR法对纯芝麻酱DNA进行芝麻蛋白2S albumim基因、花生蛋白Ara h 2基因的检测，使用酶联免疫吸附法对花生致敏蛋白Ara h3检测。实时荧光定量PCR法检测90批样品均含有芝麻成分，CT值为22.00-33.80，其中45批次还检出花生成分, CT值为25.60-38.60；酶联免疫吸附法除检出此45批次外还另检出38批次含有花生成分。通过结果比对分析及灵敏度检测研究发现，酶联免疫吸附法检出限更低。二者检测结果虽有差异，但不矛盾，均可用于纯芝麻酱中花生源检测。高含量样品检测时可选择实时荧光定量PCR法，低含量或无法预估的样品可选择酶联免疫吸附法。目前无芝麻酱中过敏原检测相关标准，本研究可作为基础研究的可靠数据并建议监管部门尽快建立相关方法，更好的规范市场行为，保障人民的饮食安全。
In this study, Real-time PCR（RT-PCR）and enzyme-linked immunosorbent assay (ELISA) were involved to detect peanut allergenic proteins Ara h 2, Ara h3 and sesame protein 2S albumim in pure sesame paste. The detection method validation including specificity, repeatability. Both methods showed good specificity and repeatability. After verifying, real-time PCR method was involved to detect the sesame Protein 2S Albumin gene and peanut Allergen Ara h 2 gene and ELISA method was involved to detect the peanut Allergen Ara h 2 protein. The sesame were detected by Real-time PCR test in 90 batches of samples(CT value 22.00-33.80), peanut were deteced by same test in 45 batches of samples(CT value 25.60-38.60). Besides the 45 batches samples, there are another 38 batches contain the ingredients of peanut by ELISA testing. By comparing and analyzing the results of two methods, it showed that the ELISA method had lower detection limit and which was more sensitive to the peanut. Although the results of the two methods were different, but they are not contradictory, both could be used for detection of peanut in pure saseme sample. The Real-time PCR method is appropriate for testing the samples which has high content peanut. On the contrary, the ELISA method is more appropriately. At present, there are no relevant standards for the detection of allergens in sesame paste. This study can be used as reliable data for basic research, and it is suggested that the regulatory department should establish relevant methods as soon as possible to standardize market behavior and ensure people's food safety.
